I did the clear autocomplete and removed the roamcache using the steps below on both affected users.
This solution worked for 5 days, user #1 now reports his nicknames are gone again.
Seems like it is just a matter of time before the remaining users are affected.
Still looking for a PERMANENT solution.

- Delete the Roam Cache from location C:\Users\User Profile\AppData\Local\Microsoft\Outlook\RoamCache
- Open Outlook. Go to File --> Options --> Mail --> Send Messages
- Click "Empty Auto-Complete List"
- Uncheck the option of "Use Auto-Complete List to suggest names"
- Close Outlook and restart it again
- Go to File --> Options --> Mail --> Send Messages
- Check the option of "Use Auto-Complete List to suggest names"
- Close Outlook and restart it again
- Create a new email message and send it.
- Close Outlook and restart.
- Verify if the entry from Step 9 is available.
